    What is a PCR test, and how does it work?

    PCR is a standard laboratory technique used in research and therapeutic settings to amplify or replicate tiny pieces of genetic material. PCR, often known as “molecular photocopying,” is very precise and sensitive. Primer sequences are used to amplify a certain DNA sequence specifically. PCR was developed in the 1980s and is currently utilized for various purposes, including DNA fingerprinting, genetic problem diagnosis, and the detection of germs and viruses. Without PCR amplification, it is almost impossible for scientists to examine isolated fragments of genetic material since molecular and genetic investigations require large volumes of DNA.

    PCR tests are a rapid and highly accurate method for diagnosing infectious illnesses and genetic alterations. The tests function by identifying the DNA or RNA of a pathogen or aberrant cells in a sample.

    DNA is the genetic substance that contains all living organisms’ instructions and information.

    RNA is an additional form of genetic material. It carries information copied from DNA and is involved in the production of proteins.

    The majority of viruses and diseases contain DNA or RNA.

    Unlike many other diagnostic methods, PCR tests can detect disease in its earliest stages. Other tests may miss early sickness symptoms because there aren’t enough viruses, bacteria, or other pathogens in the sample, or your body hasn’t had the time to produce antibodies. Antibodies are proteins the immune system produces to target foreign things, such as bacteria and viruses. PCR testing can detect the disease even when there are only trace amounts of microorganisms in the body.

    A small amount of genetic material is replicated numerous times during a PCR test. The process of copying is called amplification. If pathogens are present in the sample, amplification will make them far more visible.

    Other terms: polymerase chain reaction, reverse transcription-polymerase chain reaction, quantitative polymerase chain reaction, and real-time polymerase chain reaction.

    How do they function?

    PCR testing is employed to:

    • Identify specific infectious illnesses
    • Determine a genetic alteration that can cause disease
    • Detect minute numbers of cancer cells that conventional tests may miss.

    How do they function?

    PCR tests function by:

    • Obtaining a blood, saliva, mucus, or tissue sample.
    • The sample may also contain the DNA of a pathogen or a cancer cell.
    • The specimen is placed in a specialized machine. Polymerase, an enzyme, is added to the sample. This causes the sample to duplicate itself.

    The process of duplication is performed several times. Approximately one hour later, billions of copies are created. If a virus or pathogen is present, the system will indicate such.

    Certain viruses, notably COVID-19, consist of RNA as opposed to DNA. Before these viruses may replicate, the RNA must be converted into DNA. This is known as reverse transcription PCR (rtPCR).

    Both PCR and rtPCR detect the presence of pathogens. Quantitative PCR (qPCR) is a subtype of PCR that quantifies the number of pathogens in a sample. qPCR can be performed concurrently with PCR or rtPCR.

    What occurs throughout a PCR test?

    There are various methods for obtaining a sample for a PCR test. Common diagnostic procedures include blood testing and nose swabs.

    During a blood test, a tiny needle will be inserted into a vein in your arm to get a blood sample. After inserting the needle, a blood sample will be drawn into a test tube or vial. You may feel a slight pinch when the needle is inserted or removed. This often takes around five minutes.

    A nasal swab can be taken from the front of your nostrils (anterior nares). It may also be collected from the back of the nostrils using a nasal mid-turbinate (NMT) swab or from the nasopharynx, the upper portion of the nose, and throat. A healthcare provider may request that you perform an anterior nares test or an NMT swab on your own.

    You will begin an anterior nares examination by tilting your head backward. You or the provider will then:

    • Insert a cotton swab gently into your nostril.
    • Keep the swab in place for 10 to 15 seconds while rotating it.
    • Insert the cotton swab into your second nostril.
    • Apply the same procedure to the second nostril.
    • Take out the swab.
    • You will begin an NMT swab by tilting your head back. You or your provider will then:
    • Insert a swab into the bottom of the nostril and push until you feel resistance.
    • Sponge rotation for 15 seconds
    • Insert the cotton swab into your second nostril.
    • Apply the same procedure to the second nostril.
    • Take out the swab.
    • During a nasopharyngeal swab:
    • You will tilt back your head.
    • Your doctor will insert a cotton swab into your nostril until it reaches your nasopharynx (the upper part of your throat).
    • Your provider will rotate and remove the swab.

    Do I require any preparation for this exam?

    No specific preparations are required for a PCR test.

    There is minimal risk associated with blood testing. You may experience pain or bruises at the injection site, but most symptoms rapidly subside.

    A nasal swab may induce throat irritation or coughing. A nasopharyngeal swab may be unpleasant and induce coughing or gag reflexes. All of these effects are transient.
